Can a Roku Streaming Stick 4k Model: 3820X2 cast audio to an Apple airport express 2 Gen Model No: A1392? If yes, how do I set it up/ get it to work? They are currently both on the same network. Thanks in advance!

Hello!
You can stream video from your iPhone to your Roku stick, but you cannot stream audio from your Roku to AirPort.
An Apple TV box IS capable of doing this, so you may want to look into that.
If you want to continue using Roku, I suggest connecting the speakers to your TV with a wired connection.
