No you can not hook them together with a DisplayPort connection, because the Mac Mini is to old and the 21.5" iMac does not support Display input like a 27" iMac.
Depending on exactly what you want to do?
If you want to simply share the 21.5" iMac's Display with the Mac Mini you will need to use something like ScreenRecycler.

Or you could use a 6 to 9 pin FireWire cable and in Target Disk Mode access the Mac Mini's Hard Drive from the iMac.

Since ScreenRecycler needs a VPN connection (which in turn requires a WLAN, am i right?) I think I might just as well use the ScreenSharing option when the MacMini appears on the iMac under Shared devices.
I had hoped to be able to used a wired connection to show the MacMini on the iMac screen, because I assume that a wired connection would work much faster than one over the WLAN (again, am I right?)
Do I have the basic understanding and concepts right?
