Depending upon what country that you are in (music can't be re-downloaded in all countries) have you tried deleting it from your iTunes library and redownloading it via the Purchased link under Quick Links on the right-hand side of the iTunes store home page on your computer's iTunes ? Re-downloading.
If you aren't in a country where you can re-download music or if it re-downloads in the state then try the 'report a problem' link from your purchase history : log into your account on your computer's iTunes via Store > View My Account and you should then see a Purchase History section with a 'see all' link to the right of it ; click on that and you should see a list of your purchases ; find that song and use the 'Report a Problem' link.

I'm in the USA and iTunes states that I can download my songs again, but the Download button won't let me click on it. I bought them and downloaded (ONCE) 11 songs an hour ago. Four of the eleven don't work and I've tried everything to fix it. Guess I have to send an email, aaarrgggh.

I'm having this same issue, and so is my wife. We're finding it's not limited to iTunes songs. Some of the songs were ripped from CDs too.
I purchased a new album today and 2 of the tracks stopped short of their actual end, and iTunes skipped to the next track.
It seems to me to be an iTunes playback bug. It's been around for several iterations of iTunes 10 now. Very annoying.
I just posted the same question as well to the discussion area. If anyone offers a solution, I'll try to remember to share it.
