Do you have more than one library? If you do each time you change libraries you will need to activate PhotoStream for that library as PhotoStream can only be activated on one library at a time.
If you only have one library try the following: make a temporary, backup copy (if you don't already have a backup copy) of the library and try the following:
1 - delete the iPhoto preference file, com.apple.iPhoto.plist, that resides in your
2 - delete iPhoto's cache file, Cache.db, that is located in your

3 - launch iPhoto and try again.
NOTE: If you're moved your library from its default location in your Home/Pictures folder you will have to point iPhoto to its new location when you next open iPhoto by holding down the Option key when launching iPhoto. You'll also have to reset the iPhoto's various preferences.

Thanks Old Toad. I do have Aperture installed as well but rarely use this. I wonder if that might be using this issue?
With regard to deletion of the cache and preference files, normally that would be fine, but I think you may forgotten that the library folder is no longer visible under Lion. How do I access it?

On 10.7: Hold the option (or alt) key while clicking on the Go menu in Finder to access the User Library

Thanks Terence, that's a new on me. I will investigate and report back.
I see a lockfile in the preferences folder, in addition to the standard plist preference file for iPhoto, but as this (the lockfile) is zero bytes, I guess I should leave it?

In Lion the Library folder is now invisible. To make it permanently visible enter the following in the Terminal application window: chflags nohidden ~/Library and hit the Enter button - 10.7: Un-hide the User Library folder.
