Try resetting the device. Hold down the sleep and home keys, past when you see the red power down slider, and unitl you see it reboot. Then try again.
Are you in a situation where restrictions could have been enabled? (as in is it your ipad or does someone else control it?). Because I believe that youtube can be locked out through the restrictions part of settings.

I would put the youtube bookmark on my homescreen. Go to youtube.com in safari, touch the icon with the arrow pointing out. Add to homescreen or something like that.

I was having the same issue with my iPad 2. I did what Skydiver119 said about resetting your iPad and it worked for me

I had the same problem for no apparant reason. I just closed the app and opened it again and it worked.
