While in the Finder click this key sequence Command Shift U - this will open your Utilities Folder.
Now open System Information.
Once you have System Information open, on the left under Hardwar Click on USB, then on the right upper part of window click on Built-in iSight.
Do you see any error message in the information for your iSight?
The next thing to do is go back to the Utilities Folder and open Disk Utility. Slect your Hard Drive and then Click Repair Permssions. Let that finish. Once done I would restart the computer myself, but you can just try to use the iSight if you want.
If the camera is not broken or the wiring broken - Repairing Permissions just might solve it.
