No, this combination doesn't work. The connection method is controlled by the port on the iMac. In this case it's Thunderbolt so a Thunderbolt cable is required to do Target Display mode with this iMac. The MBair doesn't have a Thunderbolt port so it's not supported.

How about if the iMac has the a mini display port and the Macbook has the Thunderbolt port?

No, that won't work.
Thunderbolt to Thunderbolt (via thunderbolt cable only)
Mini display port to Mini display port (via Mini display port cable only)
