Yes, you can move your iTunes Library to an External Hard Drive and then access it from the iTunes Application on your Mac. For instructions see > iTunes for Mac: Moving your iTunes Media folder
Caution where caution is due: Personally I would only move my Library if it was backed up, and I had a second External Hard Drive to backup my Library on the first.iMac, Mac Mini, iPad, iPods, Mac OS X (10.6.8), (10.8), Safari, iTunes, iPhoto, etc

Make sure the EHD is Formated Mac OS Extended (journeled)...
To Operate from the EHD...
Start iTunes with the Option key held down and guide it to the new location of the library.

Yes, it's possible.
It's just a matter of copying your iTunes Library from its normal location in /user/Music/iTunes to its new location on your external drive. Then point the iTunes program to the new location of the iTunes Library.
