Are you using the same USB cable that you normally use to sync the Shuffle?

Are any other devices plugged in via USB?
And again from above:

Oct 11, 2012 12:48 PM in response to planb77by jamainehemmings,Yes, I'm plugging it straight into my mac. The cable is the original cable and I've tried others. I have my portable hard drive plugged into USB as well, but this has my iTunes library on it and its worked fine like this before.

Can you think of what may have changed on the Mac since the last time you were able to success connect and sync your Shuffle?
Maybe work through the remaining troubleshooting in this Apple support document.
