If you hold the Option key and startup into the Startup Manager, can you see the Macintosh HD..?
If you can, what happens when you select and try starting up from the Macintosh HD that way...?

Yes, I've started up using Startup Manager and saw my HD, selected it and still got the circle w/line. I've opened System Preferences while in Safe Mode and confirmed the startup disk that way, too. Same result.

If you have a (white) Retail Snow Leopard Upgrade Disc, then you should be able to Repair your Macintosh HD as per > Mac OS X - Using Disk Utility to Repair a Disk
Follow the direction for Disk Repair, not repair permissions which should only be done using Disk Utility while running in OS X.
Unfortunately if repairing the Macintosh HD as described above doesn't work, then you will have to seek help from your local Apple Store or AASP.
