In the terminal:
and to get a very detailed report on the state of your DNS cache in /var/logs/system.log:
sudo killall -INFO mDNSResponder

Unfortunately, this is an inadequate answer that does not take into account that the flushcache command worked in previous version of OS X.
As is stated in the help article I am refering to.
