How did you upgrade to Mt. Lion? Did you make a clean install and and used Migration Assistent to restore? Then your instrument library will not have been transferred., see Christoph Drösser's post:
Then the easiest way to fix this would be to find a backup and to restore the instrument library:
/Library/Application Support/GarageBand/Instrument Library/
Also check, if you installed all of the additional Mainstage2 content,
If not, disclose the Symphony Orchestra download list and force the download of the instruments again.

Thanks for your reply. This has helped me get closer to the problem.
I've checked /library/Application Support/Garageband/Instrument Library/ It looks to me like everything is there. So I go to G'band and open up a previous file that contains the software instrument "orchestra glockenspiel'. I get a search box and G'band tells me that it can't find GLS2_Pla_mf_G5.wav. I think this particular file is one of many that actually allows the Glockenspiel to play. These .wav files are what is missing. The instruments appear in the list in GarageBand, but they don't speak in the arrangement. They are silent.
This has occurred since I installed all the Mainstage2 content. I've checked the additional downloads for M2 and everything shows as 'installed' including the above mentioned Glockenspiel.
This is a curious thing because some of the Orchestra instruments speak, some don't. It appears that I have only lost some. Cello Section is another missing voice that appears in the list, but doesn't speak.
Where are these missing .wav files?

In my Library this file is called "/Library/Application Support/GarageBand/Instrument Library/Sampler/Sampler Files/Glockenspiel/Glockenspiel_Pla_mf1/GLS2_Pla_mf_G#5.wav"
with an ending "GLS2_Pla_mf_G#5.wav".
Was that a typo, or do you actually get an error message for a different filename? I have the suspicion, that Mainstage uses slightly different filenames for some of the sampler files, and if you have older oolder instruments in your library, Mainstage may not have replaced them with the newer versions.
You might try to remove the Instrument Library completely (move it somewhere else, but don't delete it) and then download the instruments again in Mainstage.
