Pressing the power button puts the iPhone in sleep mode, where the display is off but the phone is still on, "hibernating," and will come on again by simply pressing the home button or the sleep/power button.
Holding down the home button until you see "slide to power off" in red and then sliding it across the screen will completely power the phone down. In order to turn it back on you would need to hold down the power button again until you see the Apple logo, and wait for it to boot up.

No. Screen locked is Sleep mode. The phone is not off. It can still receive calls, texts, notifications, email, and updates for apps running in the background.
To turn the phone off completely hold the button on top of the phone until a "Slide to power off" message appears on the screen, then slide the control.
