Okay. Try the following troubleshooting document:

Nov 28, 2014 2:44 PM in response to ciangalby Jarek120,So I did all the updating crap and tried restarting my computer five times, and iTunes still wouldn't open.
LONG STORY SHORT––HERE'S WHAT FIXED IT:
1.) Simple update to OS X from the Apple menu>About this Mac>Software Update
2.) Download newest iTunes 12.0.1 program
3.) Restart computer
4.) Log out of your main user and sign in as guest>Open iTunes, it SHOULD open.
5.) After hitting all the "Continue" buttons, you'll get to the iTunes page we're all used to seeing, once there, sign out of guest and back to your main user.
6.) Enjoy iTunes.

Sounds like you might have a Mac. Download a fresh copy of iTunes and install it from http://www.apple.com/itunes/download/

Jul 23, 2015 3:14 AM in response to turingtest2by CupOJoeGA,I am having the same problem on my Mac (Yosemite 10.10.3). click on iTunes after most recent update....nothing. No bouncing icon, no launch, nothing. I have restarted several times, logged in as a guest user, and even re-installed the latest version (did not uninstall, just installed over what was there). Any other ideas? Terrible timing,of course, because I needed to use iTunes for a specific purpose before going to work this morning and this is the first time it has failed to open. Help!

Jul 23, 2015 6:58 AM in response to ciangalby princewuz,Hi, I've been having a few difficulties with iTunes for a few months. I updated it to the most recent update, but the problems still persist:
1. Every other time I open iTunes, it crashes.
2. It wont connect to the internet/iTunes Store
3. It wont read my iPad although the iPad comes up on windows explorer (and other computers).
I've looked at various ways to sort this online, but none have helped so far.
Does anyone know how to fix the above problems?

The steps in the second box are a guide to removing everything related to iTunes and then rebuilding it which is often a good starting point unless the symptoms indicate a more specific approach.
Review the other boxes and the list of support documents further down the page in case one of them applies.
The further information area has direct links to the current and recent builds in case you have problems downloading, need to revert to an older version or want to try the iTunes for Windows (64-bit - for older video cards) release as a workaround for installation or performance issues, or compatibility with QuickTime or third party software.
Your library and device backups should be unaffected by these steps but there are also links to backup and recovery advice should it be needed.
If the issue persists and you have a 64-bit system try the for older video cards version which is a 64-bit installer for mostly 32-bit code, similar to all 64-bit versions of iTunes up to 12.1.
