There are two articles I would suggest you read over and work through to begin the isolation and troubleshooting process.
If your voice is too faint or sounds unclear using iPhone, or iPod touch - Apple Support
Because the other party cannot hear you and you cannot hear them while on a call, the articles above help to troubleshoot both the microphone and the receiver. This will also help us isolate the issue to being system wide across your iPhone or just occurring while on calls.
I know situations that arise after an update can be very frustrating, but we are committed to making sure you are happy using your iPhone with iOS 9. Please reply to let us know if the above steps resolved this situation or if it continues so that we can continue troubleshooting.

Sep 24, 2015 1:05 PM in response to daverpigby hvpia239,I was having the same issue until I shut my phone off and then powered it back on. It has worked for a lot of people. I also have an iPhone 6 that I just update to iOS 9. Hope it works for you.

Sep 28, 2015 9:55 AM in response to daverpigby TheBigVanMan,My wife has this same issue after upgrading to IOS 9.0 on a 5S (Sprint is the carrier). Last night, I installed 9.0.1 and it still hasn't fixed it. I believe this is a software issue and not a hardware issue since I tested with some alternate microphone sources including one hardwired (plugged in headphones with a mic) and one via bluetooth (connected a headset). When this problem was happening, no microphones could pick up my voice (tested using the voice recorder app).
After a restart (holding down the sleep and power buttons), microphone and receiver function were restored, but not for long. This morning, the same problem is back and a restart or power-off did not correct it. Anyone else have any other suggestions aside from downgrading or waiting for a fix?
