It sounds like you are unable to use the speaker on your phone because it seems to think that headphones are plugged in even after having cleaned out the port. I would next try plugging and unplugging a pair of headphones a few times and retesting the issue. If the issue persists, I would use these steps specifically from the following article:
iPhone: No sound or distorted sound from speaker

- If the iPhone is paired with a Bluetooth headset or car kit:
- Try turning off Bluetooth.
- If you experience difficulties with the Bluetooth feature, follow these troubleshooting steps.
- Restart the iPhone.
- If restarting doesn't fix the issue, ensure that your iPhone is updated to the latest version of iOS.
- If the issue is not resolved, please contact Apple Support.

Sep 25, 2015 1:29 AM in response to Schmitycby lanastiel,I have the iPhone 6, and came across this question after my own phone became stuck on headphone mode.
Hard reset, using a rolled up tissue to rid the jack of debris, and wiggling the earphones around helped fix the issue.
I also went to sounds and played my ringtone. Low and behold, the speaker was fine, however it was still stuck when using applications or playing music. I reset my phone again and it fixed the problem
