I'd recommend a locally connected external drive as your primary library with the NAS as a backup location. Select a track that won't play correctly and press Ctrl+I to get info. What do you see on the file tab for the location of the file? Is that what you would expect to see in terms of the artist, album, and track properties? What file really plays? Is that file also broken? Do any of the properties of tracks change when the wrong audio is played?

However, I have been using itunes with my Iomega NAS for a few years now without problems. My main reason is that I also have a DLNA Amp that is fed I believe by Twonky installed somewhere withing the Iomega software on the drive. Also the Iomega drive is designed so they say to work with itunes.
My thoughts really turned to some corruption within the itunes database or rather how it was being read.
Ironically I turned it on this morning and everything is working fine, so perhaps it is a W10 network problem rather than itunes.
Anyway your Ctrl+I suggestion next time it happens will perhaps shed more light on the problem.
