Q: In a daisy-chain of two Thunderbolt Displays, could i replace the first Display with an (e.g.) 2012 iMac and use Target Display Mode to power both?
If my Thunderbolt Displays have the ability to be daisy-chained and get their signal over a single Thunderbolt cable, shouldn't an iMac in Target Display Mode also have this ability? Is an iMac in Target Display Mode capable of simulating a Thunderbolt Display not only to get a video signal for itself but also for a Thunderbolt Display which is connected to it?
I have heard that some people used their iMac as a Target Display with Target Display Mode while the iMac was connected to another Monitor (Not a Thunderbolt Display).
The result was that the iMac's internal Display switched to the Target Display Mode, but the connected Monitor didn't.
I haven't yet found someone who tried this out and published his experience with a Thunderbolt Display connected to the iMac.

Unfortunately, a bit more research would have answered and has answered my question.
Yes, I can replace one of the Thunderbolt Displays in the chain with an iMac. The replaced Display has to be the one which is not directly connected to the source mac.
