Have you tried holding the home button and then tap the power button or holding the power button and than tap the home button? Sometimes one of those methods is more effective than holding both buttons down at the same time.
Are you having any issues with either of the two buttons involved?
Describe how you are trying to take the screenshot and explain what happens when you try it.

Feb 21, 2016 5:00 PM in response to LaurieJSby Demo,Try a forced restart. You will not lose any data. Some people refer to this as a reboot. It can clear up weird unexplained issues.
To force restart your device, press and hold both the Sleep/Wake and Home buttons for at least ten seconds, until you see the Apple logo.

Hi LaurieJS,
If you're having problems taking screenshots on your iOS device, please try the following steps. Try each step, then see if your problem still persists. If the problem does not persist anymore, you can stop. If the problem persists, continue following all the steps until you've completed all of them or you've went through all of the steps.
- Make sure you're running the latest iOS version
- If you aren't sure if you're running the latest iOS version, check out this support article from Apple:

- Make sure the app you're using let's you take screenshots.
- Make sure no accessories are blocking or interfering with your ability to use the home button and power button to take a screenshot.
- Try Resetting All Settings
- Open Settings > Tap General > Tap Reset > Tap Reset All Settings
- If you're asked for a passcode, go ahead and enter it.
- Then tap on Reset All Settings again to confirm.
- Open Settings > Tap General > Tap Reset > Tap Reset All Settings
- If the following steps didn't help you can enable AssistiveTouch to take screenshots.
- Open Settings > Tap General > Tap Accessibility > Tap on AssistiveTouch > Toggle AssistiveTouch On.
- The AssistiveTouch menu will appear on your screen.
- Tap on Device > More > Screenshot.
- The AssistiveTouch menu will appear on your screen.
- Open Settings > Tap General > Tap Accessibility > Tap on AssistiveTouch > Toggle AssistiveTouch On.
I hope these steps help you get back to being able to taking screenshots. At the very least, if you're not able too you can at least use AssistiveTouch to help you take screenshots.

Feb 22, 2016 6:30 AM in response to Demoby LaurieJS,No, no, I appreciate your help!!! Literally nothing happens when I try to take a screenshot. Pressing the home and lock button, alternating buttons etc. nothing happens except then if I press too long my phone turns off!

Feb 22, 2016 6:43 AM in response to LaurieJSby Demo,Both buttons must be working properly or you would not be able to take a photo using the home button or turn the power off using the power/Sleep button. If you tried resetting all settings as was suggested by Coolbobcat2, the next thing that I would try is to backup the phone, restore to factory settings and restore the backup. If that fails, I'm stumped. The only other thing that I can think of would be to restore to factory settings.
