I understand you are unable to backup to iCloud. I would need more information on what exact behavior you are experiencing to provide a better answer. Please include any error messages received. However, here is the best troubleshooting guide to go through for issues backing up your iOS device to iCloud.

Besides jason269's suggestion, this topic is being discussed a lot in the "iCloud on iOS device" forum. There have been several comments ranging from toggling automated backups off & on, signing out of and into iCloud, to it being a problem on the Apple server with the iOS 9.2.1 upgrade. I don't know. I was having a problem with automated backups to iCloud until recently and then it somehow seemed to fix itself. I don't know how - I did do a complete backup and restore for another reason and maybe that had something to do with it.
Are you able to do a manual backup to iCloud? I was always able to do that. Some folks weren't. As a precaution you should at least backup to iTunes until you get it working.
