I understand one of your songs isn't behaving as you'd expect. You're in the right place for help - I know it's important to be able to play your content as expected.
In this case, it may be a good idea to import or download the song again. Then, try to play it using iTunes or import it onto your iPhone 6s again.
If this doesn't resolve the issue, what happens when you attempt to play the song? Does audio play? You may also consider updating iTunes on your computer, if you haven't already:
Get the latest version of iTunes
If you continue to see the same behavior, be sure to include in your reply if the song was purchased from the iTunes Store, or if it came from another source such as a CD.
