Have you tried starting Mail while holding down the Shift key?

Another way to try to get Mail to open clean would be to delete Mail's saved application state.
You can pretty much wipe out all of Mail's settings by deleting the Container which can be found in your user Library/Containers folder. It is a folder named com.apple.mail
