What happens when you try?
Do you get an error?
Does the iPhone not show up in iTunes?
Do you not have iTunes installed?
What exactly is happening?
Updating should not delete any content from the iPhone as long as it completes correctly. But its always always a good idea to backup either to iTunes or iCloud before updating in case something does happen.

That's a useful info. "can't seem to backup" (or some say "does not work") is too vague and we don't even know what happens when you try. Anyway, did you ever successfully get iTunes to "talk" to your phone? What iTunes version are you using? Have you tried connecting the phone to a different USB port on the PC? When you plug in the phone, take a look at the phone display for a prompt, if there's any.
