While deleting caches is normally safe, see Myth #4.
Note that your Library folder contains all of you Mail and attachments. If you've got a lot of email with large attachments, you can really fill up the Library folder.
Your user installed Fonts and Log files are also in there. If you have a program that is constantly writing log entries without cleaning it up, it could create a large file after a short time.
The Cache folder on my system is 1.22 GB with the total Library of 2 GB.
Everything else in there shouldn't be taking up much space.
All the above responses are largely true, but not always for every situation.
Most likely, you've either got lots of old junk laying around, or you just need more space, like on an external FireWire drive. Still, 25gb in your Library folder is huge.
I use a free program called OnyX at http://www.titanium.free.fr/index_us.html
You'll find numerous favorable references to it in these forums. It will do lots of maintenance and clean-up routines.
There are other, similar programs as well, some free, some not.
