I had exactly the same -15000 error with iTunes 8 on 10.5.4 (fully updated) with 7.3.2 in the Express.
I opened the firewall port for iTunes using the 'Set access for specific services and applications' and adding iTunes and 'Allow incoming connections'... then restarted iTunes and it immediately burst into life.
I don't like opening up my machine to any and all UDP traffic as a way to solve this issue. **** it, it worked with UDP shutdown with V7, it should work that way with V8.
Also, I'm tiring of iTunes updates horking all of my settings and preferences. Also, the lack of preference settings to get rid of the Genre setting is a bit unnerving too. I have disabled it with the 'defaults write com.apple.itunes show-genre-when-browsing -bool FALSE' command but I should not have to resort to 'escape to terminal' to do a simple preference setting.
I had this same problem I fixed it by turning off my firewall on my Vista Machine that has the ITunes program on it. After you turn off your firewall then shut down ITunes and then turn it on again seems to work ok. I would feel better if I could leave my firewall on but what can I do the love to use my Airport Express outside by the pool. Another Item that Mr. Jobs released before it was ready. Just think of all the Bad reviews that would have come out if this was done by Microsoft......Hello Apple Fan Boys

Again I have a Vista Machine. I turn off the firewall in Vista and all worked well but I wanted it to also be on. So I rebooted my Vista computer and then turn on the firewall and now my airport express works great with the firewall on and my IPhone is controlling my airport express with the remote program for the IPhone...
I just did a firmware update to my Airport Express from 6.2 to 6.3 (very easy with update from Apple) and it solved the problem of connecting to my remote speakers via Airtunes. Thanks for directing me to this solution guys.
If firewall is good, and the firmware is up to date, try renaming the network, the base station and the itunes speakers in the airport utility. that worked for me
I had the same error -3256 problem. Fiddling with the Firewall did nothing. HOWEVER, I then turned on Configure IPv6 Automatically in the Network System Preferences for the Airport (under TCP/IP in the Advanced settings) and applied the change. That solved the problem. To verify, I turned off IPv6 and got the -3256 problem again. Re-enabled IPv6 and Airtunes once again worked.
Of course, your mileage may vary. It always does. . . .
