Why won't Garageband pick up any microphones?

Hi,
I need to record a sample for some bands and Garageband seems to be the way to do it. I have managed to get my guitar to work with it once before, which was a chore to begin with, but now I have the same problem, but worse!
My new microphone isn't being detected at all (neither is a different one), and I've tried everything in the help menus.
I checked the volumes, changed input source, etc.
Can anyone help me get my Mac to realise there's a Mic in the Line in port?

Thanks

Macbook, Mac OS X (10.5.5)
