Do I need a Router/AirPort Extreme Base Station with Gigabit Ethernet to make both the machines speak.
No. If you'll be satisfied with a wired connection, you can simply connect their Ethernet ports together. If you prefer a wireless connection, you can connect them via their built-in AirPort cards. [AirPort 1.3: Using Computer-to-Computer Mode (IBSS)|http://support.apple.com/kb/TA28186] describes how to do this. The connection will be slower than Gigabit Ethernet, but unless you're moving lots of big files routinely, the difference may not matter.
More over do I need a AirPort Extreme Base Station with Gigabit Ethernet to send music form itunes of both the machines library to my music system.
If you want to stream iTunes to your stereo using one of the stereo's audio input jacks, you'll want an AirPort Express (AX). An AirPort Extreme (AE) has no audio output jack.
I want to use migration assistant to transfer files and contacts from mini to notebook. Will it work via AirPort Express Base Station with 802.11n
Yes, but the direct computer-to-computer options I've described would suffice. [MacBook (13-inch, Aluminum, Late 2008) and MacBook Pro (15-inch, Late 2008): Migration Tips and Tricks|http://support.apple.com/kb/HT3231] describes how to migrate wirelessly and using Ethernet. Ethernet would be significantly faster for you.
