Try this. Remove the Genius database. In the "Finder" go to the /username/Music/iTunes/ directory and delete the "iTunes Library Genius.itdb" file. This should allow the program to launch again. This file will be recreated upon first use of the "Genius" feature.

thank you SOOOO much! I am about to go on a rather long plane ride and was NOT looking forward to not having my iPod up to date with my iTunes. All the sudden I get this error messages and I'm freaking out...you saved me a massive headache. I owe you lunch.
Hi, I have been unable to open my iTunes for a few days now. I have tried delelting the genius file, nothing. I deleted other library files and genius files, nothing. Uninstalled it a few times and reinstalled. No help from Apple. Does anyone know what I should do. I have windows vista if that helps.
I had this same problem but removing itunes genius alone didn't work.
There was another file called itunes extras. When I also removed it, I was able to open itunes.
