Is this a USB mouse? If so, try a different USB port and see if it makes a difference.

Have you tried restarting the iMac?

Have you tried booting in Safe Mode? http://support.apple.com/kb/HT1455

I'm not certain if Safe Mode can help with this but it won't hurt to try.

Resolve Startup Resolve Issues and perform disk maintenance with Disk Utility and fsck

Do you know if you have "mouse keys" enabled in the Universal Access preference pane? If so, you could try and use the numeric keypad on your keyboard to move the mouse. If you have the new keyboard without keypad, use fn + number keys to move the mouse. 2, 4, 6, 8 is for pointer movement, and 5 is for doing a mouse click.

Might help in identifying if it is an hard- or software-issue.

Can you boot from your install disk?

Try the keyboard commands that rockstar0815 provided.

Those keys only work with Mouse Keys turned on thought right? OP can't check System Preferences.

Restart and hold down the mouse at the same time to eject a disk.

1. Restart the computer.
2. Immediately after the system startup tone, press and hold the Command-Option-O-F key combination. The computer starts up to a text-only screen, indicating that you are in Open Firmware.
3. At the prompt, type: eject cd
4. Press Return.
5. Type: mac-boot
6. Press Return

Maybe that will work.
