Because you have already tried reinstalling Skype and
everything else already suggested here, consider these possible remedies for your different problem:

(1) You do not post which Mac and Mac OS you are using, so I cannot tell you exactly how to do this, but, if you need it, you can get assistance via your Mac's Help menus.
Start by checking that your Mac's
System Preferences ... > Accounts > Login Items do not automatically launch any apps that use your iSight. If you use any widgets that access iSight, be sure not to launch them before trying to use Skype either.
While it is certainly
possible to have multiple applications that can use iSight
open at the same time, as explained in Apple's
iSight "Your camera is in use by another application" message

iSight can be
used by only one application at a time.
Applications, login items, or widgets that are already using your Mac's iSight, whether in your current or a different user account, will give you the "in use" or "no camera" error. If you cannot find the conflict, shut down your Mac and power up again to resolve this problem.
(2) When you shut down (do NOT merely restart) your Mac, you should also shut down your modem and, if you use one, your router. Wait about thirty seconds and restart the modem. Wait for its self-test to complete. If you need help, see the users guides that came with your equipment. Then, restart your router and allow its self-test to run to completion. Finally, restart your Mac. Then, with no widgets or other apps that can use your iSight running, launch Skype and see if your problem persists.
(3) If the problem still exists after restarting, try every relevant suggestion re: your iSight from Apple's article entitled "How to Troubleshoot iSight."

The reset suggested there often resolves problems like yours. If not, it may help you identify the source of the problem so you can take the required actions to get your system working properly.
I have tried everything that other people have tried and it is still not working for me.
(4) If your problem is not resolved by any help you get here or from the Skype forum, your Mac may have a hardware problem.
If after carefully following all relevant suggestions in the "How to Troubleshoot iSight" article, your iSight still does not work, and if your iSight is not recognized in your Mac's
System Profiler, yours is most likely a hardware problem. In that case, contact Apple or an Apple-Authorized Service Provider for service.
