Your company could well be blocking the connection, if you're on the company network. Before you go through a lot of troubleshooting, I'd suggest you ask your IT department about it. That might save you a lot of unnecessary work if they are indeed blocking.
If they say they're not blocking the iTunes Store, or you have the computer at home, not at work, post back and we'll take it from there. when you post back, please mention the version of iTunes you're running, and how you are connecting to the Internet.
