The Alpha tool, which I haven't had to use in a while, but now desperately need, appears to have been replaced by a Sketch tool. Does anyone know what happened to it, and how I can access it again? Thanks in advance.

The Instant Alpha tool should be right next to the Sketch Tool. Looks like a Magic Wand.

I believe you. There is something hinky about your copy of Preview. It looks like you have two Sketch tool icons and no idea what the "Show Markup Toolbar" ,"label/button?" is doing there.
