The shortcuts for screenshots are not working. I hear the camera shutter sound but they are not appearing on my desktop and when I search for any images created today on my Mac, they are not there.
Did you checked your System Preferences>Keyboard>Keyboard Shortcuts>Screen Shots settings to see if it is enabled? We have also one that I like that is not on that list and works here, SHIFT

Interesting, the shortcut you mentioned SHIFT
COMMAND+4SPACE BAR works but the others still don't work. I checked my preferences and all the screen shot tick boxes are clicked on.
I've been hunting through forums and I found a response to a similar post for 10.5.7 which says:
"Navigate to Macintosh HD > Users > username > Library > Preferences and find the preference file "com.apple.ImageCaptureApp.plist". Move that file to your Desktop and restart your Mac. If the screen capture function returns, delete the file."
Problem is that I don't seem to have that preference file. Is that something that changed in 10.6 or could that be part of the problem?
I have this property list with a different name, com.apple.Image_Capture.plist, you could try to change those settings for shortcuts and also search in that panel for some other entry using the same shortcut. If no other entry use it then just change to test. First change the combination keys, close System Preferences and test, if you still don't get it working try logout and login again, if still get nothing try restarting, then if you can't use it yet try to find that plist and post it's contents here.

Adding. I just found another plist file named com.apple.symbolichotkeys.plist that probably is the right configuration one.
