Is it ok to install three ram sticks?

I have a late 2015 5K 27" iMac... I've been considering upgrading the ram, but I'm not sure I need 8GB more right now (for a total of 16), and was thinking of just getting another 4GB stick.

Is it ok to only use 3/4 ram slots, or will that result in latency issues?

That works, but only in single-channel mode instead of the dual-channel mode you get with matched pairs.

There are no latency issues, but single channel memory has up to 50% lower bandwidth that usually results in ~5% slower overall performance. I recommend sticking with match pairs.
