If you mean 2 Apple Watches, 2 users and 1 iPhone, then no. While you can pair multiple Apple Watches to an iPhone, ONLY 1 can be active. This is for the user that collects watches for different occasions and wishes to switch the watch they are wearing for work, play, entertaining, etc...

Basically 1 iPhone per Apple Watch user is required.

If you mean can you put your Apple Watch on your Wife's wrist and take her ECG, then sure. It will be stored on your iPhone and you will need some way to keep track of which reading is yours and which is your Wife's, but nothing stops you from putting the Apple Watch on someone else's wrist for a 30 second ECG reading.
