Apple Mail does not directly support baseline (sub/superscript) functionality, so you will have to compose the appropriate string in either TextEdit (Rich Text Document) using the Format menu : Font : Baseline menu, or in Pages with the ^⌘-minus or ^⌘-plus signs to effect subscript or superscript respectively. Then copy/paste into your Mail compose body.
As John Galt has mentioned, your Mail preferences : Compose : Message Format should be set to Rich Text before the first paragraph will work.

A subscript, as in text? You need to compose the email message using Rich Text or HTML, and use the corresponding markup tools.
