I want to know what "Back Up Automatically" in "System Preferences".
If I turn off "Back Up Automatically" and click "Back Up Now" in the menu bar, is it OK?

It’s up to you. You can choose what you prefer. If you enable ‘Back up automatically’, TimeMachine will start every two hours with backing up what has changed on your mac. If you don’t want that: leave ‘back up automatically’ unchecked and don’t forget to back up regularly

Time Machine is busy with encrypting the backup disk. That can take a very long time. Just be patient (at the screenshot, you can see that 54% is ready) and wait until encryption is finished.
Three folders and one 'Latest' is normal. It will help you to go back to an earlier backup if necessary.

Thank you very much for your help.
Today, I ran Time Machine on external HDD.
I turned Mac into Sleep mode with the external HDD connected to Mac.
I've found out "three folders and Latest", as seen in the screenshot below.
If I want to stop Time Machine now, how would I do?
Time Machine looks like going indefinitely.
