I recently connected my AirPort Express to an amplifier through optical out and noticed that there is a time lag of more than 2 seconds when streaming audio from MacBook Pro 2019. However when I stream from iPhone, the time lag is much less, almost unnoticable.

You are right. Music app works without a delay. But I want to watch video from the internet, where my TV is connected through HDMI and audio is connected through AirPort directly to my stereo. When I stream video from a browser, there is this audio lag. It is strange that some applications cannot work with AirPlay 2. I would expect this to be a system property, transparent for applications.
If you can download the videos, you could import them into the Music Video section of Music, and play them from there.
You could also connect the headphone output of your Mac or TV to the stereo amplifier.
The Mac may be using AirPlay, not AirPlay 2. The older AirPlay has a delay. Some apps on the Mac may not support AirPlay 2. Try the Music app.
