If I have uploaded all my photos to my iMac, can I delete them from my phone and still have them on my computer?

If you have iCloud Photos enabled on your Mac, the photos will be deleted there too.

If you disable iCloud Photos on your Mac, you can retain them there. Try a few, as a test.
