There are no built in tools in Keynote for Mac to annotate when Keynote is in Play.
The two workaround methods are
1 - use an iPhone or iPad with the Remote Control function: https://support.apple.com/en-gb/guide/keynote/tan85f5c958c/mac
2 - use a third party application such as ink2go from the Mac app store (http://ink2go.org)
I'm pretty sure this doesn't exist in Keynote yet - it doesn't even show up in Apple's Keynote help page.
Here's a workaround using your MacBook Pro and Zoom:
If you're using Keynote 10.1, just release last week, you can play a windowed Keynote version inside Zoom (Play menu > Play Slideshow in Window). Start your presentation in Keynote. Then choose Play in Slideshow in Window. Next, launch Zoom and choose Share Screen, and select your Keynote presentation. Now your Keynote presentation is shared, and you can use Zoom's annotation features, which work very well.
