How do I lock my phone

If you are using the Backup now option you don't need to (and cannot) lock it.

If you are wanting it to automatically backup while charging and connected to wi-fi, either press the right side button or do what I do -- just let it lock itself automatically based on the time chosen in Settings > Display > Auot-Lock.
