How do I fix "Disk Utility First Aid can't repair external volume 'The repair cannot be performed because one or more volumes are mounted. : (-69565)'"

In Finder, try to eject all of the Volumes on the drive. If it gives an error, try to resolve that problem.
For instance, if it can't be ejected because something is using it, make sure all files on the drive are closed.

The volume was a clone backup of my main HD; the Mac may have booted from it, since the main HD was was troubled.

What drive or volume are you trying to repair? If it's one of the two Catalina volumes you must be booted into the Recovery volume (boot they the Command+R keys held down) in order to rum first aid on the drive.
This is an external clone backup of my Macintosh HD, so has a bootable version of Catalina on it. I was able to test/repair it from Recovery mode.
