Markups will not save.

There is known bug in iPadOS14 that affects saving of markup documents/photos. There is a workaround until it’s fixed; simply trim/resize the image before saving.

Are you having this problem with edits in general (as in your text) or with markups in particular (as in your title)? We have seen suggestions that markups without other edits may get lost in recent updates. The suggestion is to make or undo some minor edit before doing the markup.
Resizing or trimming before saving/sending works.

I agree that saving and sending the markup is critical. I just verified that my iPad Pro 12.9-inch 2nd generation (14.2) and my iPhone 11 Pro Max (14.2) can each do markup on a recent photo, save it, and send it by email with the markup visible. Now we have to figure out what is different about your library, your devices, or the photos you are marking up.
Are the photos you are marking up old or new? Mine were less than a hour old.
Had the photos you are marking up been edited since they were taken? Mine had not. (One had multiple markups but not other editing.)
Are your photos JPEG, PNG, or HEIC? Mine were one HEIC taken with my iPhone and one PNG screenshot from my iPad.
I was juggling the same screen shot among the iPhone, the iPad, and a Mac. They mostly tracked one another (in real time on the local network), but a couple of the changes got lost along the way. Something is not quite right there.

I recommend that you submit your experience of the bug to Apple using the Product Feedback page - as issues are not necessarily afforded priority unless sufficient reports are seen by Apple.
Please record your feedback as a bug - and be sure to include details of your iPad model and installed version of iPadOS in the main body of your report.
Markups cannot be saved. It doesn’t matter which iPad I use, or my iPhone. I just get an error message. I never had this problem before the upgrades. I use the Markup feature daily.
The markups themselves are not the problem... I just can’t save or send them, which is pretty much the point.
