What settings will prevent iMac 2020 running 11.0.1 from going to sleep during long upload? Every setting combination in System Preferences > Energy I have tried does not work.

I don't have an iMac, but on my MBP:
(you could let your display turn off).
Then you need to see if there is something scheduled (like your mac being set to sleep or shut down some time during the night):

There is a setting to logout after a period of inactivity, maybe that is what is taking place. I will have to check it in my mac, but I think it is System Preferences->Security & Privacy->Privacy
It is in System Preferences->Security & Privacy->Privacy, indeed.
Click the padlock to authenticate, then click Advanced...
and make sure that the first box is NOT checked.
Switching OFF the "Log out after..." switch located; System Preferences > Security and Privacy > Advanced, did resolve my issue, see "Dec 5, 2020 2:02 PM in response to Luis Sequeira1" above. Also it worked fine with System Preferences > Energy Saver setting set to default.

This is how I have my iMac setup to not sleep:
Are you sure that the mac is going to sleep and not just the screen?
Upload does not progress, and resumes automatically upon login. I also tried setting preventing screen from sleeping, Can you screen shot settings you use when not on battery.?
My "Log out after..." switch is now off. I am trying this, with default Energy Saving settings and let you know – Thanks
