I have an early 2015 MacBook with USBC port and a 27 inch late 2009 iMac. The iMac doesn't have a thunderbolt port, but has a mini display port. Is it possible to connect them to make the iMac a target display monitor?

The Late 2009 27" iMac supports Target Display Mode using a Mini DisplayPort cable.
To make it work, you need a USB-C to Mini DisplayPort adapter cable.

Note that, although your current hardware configuration supports TDM, that can cease if you upgrade the MacBook Pro to Big Sur. Because the TDM article is archived, it isn't updated, but reports here say TDM is not working with Bg Sur on the SOURCE computer even if the other hardware requirements are met.
