How do I set up my iPad to back up if it has to be locked?

If the iPad is locked/disabled, you’ll not be able to create a new backup of the device. If the iPad is disabled, all opportunity to backup has been lost.
If your iPad is disabled, or you have forgotten your iPad passcode, you’ll need to follow this process:

You’ll need access to either a PC (with iTunes installed) or a Mac (with iTunes or Finder - as appropriate for the installed version of MacOS).

Unless you AppleID account has not been fully/correctly configured, or has been seriously neglected, recovery of the AppleID and password should not be difficult.

When you recover access to your credentials, you would be well advised to log-in to your AppleID account from a web browser - and verify/update any email addresses and trusted telephone numbers:
If your iPad is configured to use iCloud backup, it will do so automatically each day - so long as the iPad is connected to both WiFi and its Power Adapter.
If the iPad is not already configured to use iCloud backup - and the iPad is locked and you cannot remember your passcode - then it is impossible to initiate a manual backup. In such situation, whilst the iPad is locked, all data will remain securely encrypted.
