With an inadvertent click of the mouse, my Downloads folder has disappeared from the dock and despite opening the Downloads in finder and dragging back to the dock, it has refused to move! Can anyone help please?

Dragging "Downloads" from the Finder Sidebar should work.
Following your 'Add to Dock' suggestion and using Control Mouse click I have managed to add it back on to the Dock. Looks slightly different than the fan style of presentation but at least it's there now so thank you!
Now it is in Dock >> Again Right Click the Downloads folder in the Dock. Again, popup menu will appear and tick off the options desired.

Check Finder > Preferences… > Sidebar to see if "Downloads" is marked.
