Can my wife use my MacBook pro to sign into her separate iCloud account and then access and edit her full photo library (of 50,000 photos?)

Yes, simply create a new user so you can both be signed in with your own Apple ID account. Here is an article that may help.

A single Apple ID and password is signed in Mac ( that is of yours ) , with your permissions if your Apple ID is signed out from Mac system preferences and her Apple ID is signed in Mac and photo library and its preferences is established https://support.apple.com/en-in/HT204264

Then she can edit the photos .
The efficient way is - as your Apple ID and password is different than her and used on different Macs , but on the same network .
She can create albums https://support.apple.com/en-in/guide/photos/pht6d60a1f1/mac with different names on her Mac add few photos in it then share the albums with your Mac https://support.apple.com/en-in/guide/photos/pht884a8908/4.0/mac/10.14 ( you have to accept the invitation in mail and subscribe it ) she must remember the name of albums so the photos of her can be edited https://support.apple.com/en-in/guide/photos/pht5f6df5f0/4.0/mac/10.14 in your Mac .
Note : there are more than fifty thousand photos few photos are added in a single album , once the album is shared let the process of sharing be completed , then create second , third ...album and share second , then third ..... as lot of network speed is required .
