Apple doesn't directly support having iOS device backup data on an external drive. If you've followed advice such as Relocate iOS device backups to put your backup data on an external then it should be seamless as far as iTunes is concerned. Both backing up and restore take place through iTunes with no additional steps required. See Restore your iPhone, iPad, or iPod touch from a backup - Apple Support.

If, on the other hand, you have copied over a backup from another computer and now hope to be able to restore that backup from a new one, but cannot see the backup you want to restore, see this advice from Archive iPhone backup in Windows - Apple Community:

iTunes may not catalog all of the backup sets in the backup folder. If you need to restore a backup that iTunes doesn't detect you can potentially create a new one, then swap the folder names around so that the backup you want to restore has the folder name of the newly generated backup.
