I am trying to get all my photos in one place on my Mac. So I've transferred all my pics from 'Pictures' to 'Photos'. Can I now delete all the ones in 'Pictures' or will it delete them in Photos as well?

As long as you have the "copy items to photo library" checked in preferences, and you have imported the images, then yes you can delete the files from the folder, as they have been copied into the library. To be certain this works, delete them, then check they are still in photos before emptying the bin.

You could also create a smart album with ‘photo is referenced’ to make sure everything is actually copied into the library.

I backup in iCloud and on an external hard drive but just want to tidy up. Lockdown project!
The screen Tony has shown is the preferences screen. Press CMD and , [COMMA] or you can find it from the File menubar at the top of the screen.
